Comparative Evaluation of Fixatives for Immunocytochemical Staining in Diagnostic Cytology

Abstract
Immunocytochemical staining is now a important tool for tumor typing in diagnostic cytology. However it is less used than immunocytochemical staining. This may be due to methodologic difficuties. Therefore, this study compared few. fixative methods for immunocytochemical staining in diagnostic cytology. Three commonly used fixative methods were evaluated: (1) 0.05% glutaraldehyde at RT for lmin or 15min; (2) 95 % alcohol at RT for 30min, and at 4¨¬C for overnight; (3) acetone/alcohol mixture at RT 30min and 4¨¬C for overnight. Specimens were obtained from colon adenocarcinoma and lymph nodes. Immunocytochemical staining was performed with antibodies directed to 3 different types of antigens: (1) antibodies directed to the imtermediate filament proteins, cytokeratin and vimentin; (2) antibodies directed to the cytoplasmic proteins (lymphoreticular system markers), leukocyte common antigens (LCA); (3) and antibodies directed to the nocofetal antigens (carcino embryonic antigen; CEA). The reults showed that immunocytochemical staining on the specimens fixed with 0.05% glutaraldehyde at RT was better than those with other fixative methods.
